Le sindromi oto-palato-digitale distinte in due forme, tipo 1 e 2, in base alla gravità , sono malattie legate all'X dovute a mutazioni del gene FLNA. Esse fanno parte, insieme alla sindrome di Melnick Needles e alla osteodisplasia fronto-metafisaria, dello spettro fenotipico della Osteodisplasia fronto-oto-palato-digitale.
Sono caratterizzate da:
Displasia scheletrica: ossa lunghe curve, agenesia della fibula, aspetto a ventaglio delle dita dei piedi, polidattilia;
anomalie del SNC: idrocefalia, ipoplasia cerebellare, raramente encefalocele e mielomeningocele.
